Conversion of a D-Alanyl-D-Alanine peptidase into a β -lactamase.
DD-peptidases, also called PBPs (Penicillin Binding Proteins), are part of an enzyme family involved in the synthesis and modification of the peptidoglycan of the bacterial cell wall. β-lactam antibiotics target these PBPs by forming stable acylenzymes, which inhibit them in a permanent manner. This inhibition kills the bacteria. Some bacteria have acquired β-lactam resistance by expressing enzymes called β-lactamases that hydrolyses antibiotics before they reach their target. β -lactamases have likely emerged during evolution from PBP-like ancestors.
Although the conversion of a PBP into a beta-lactamase seems a priori straightforward, it has proven to be extremely difficult in the lab.
