I use the SICLOPP (Split Intein\u2013mediated Circular Ligation of Peptides and Proteins) technology coupled with a co-translational export peptide to produce large libraries of peptides in the periplasm of <em>E. coli<\/em>. Those libraries can then be mined to look for targets of interest, for example inhibitors of the enzymes involved in resistance to carbapenemases, or inhibitors of the cell wall biogenesis of Gram- bacterias.<\/p>\n\n\n\n<figure class=\"wp-block-image\"><img loading=\"lazy\" decoding=\"async\" width=\"963\" height=\"407\" src=\"https:\/\/sites.uclouvain.be\/soumillionlab\/wp-content\/uploads\/2018\/10\/siclopp7.png\" alt=\"\" class=\"wp-image-1653\" srcset=\"https:\/\/sites.uclouvain.be\/soumillionlab\/wp-content\/uploads\/2018\/10\/siclopp7.png 963w, https:\/\/sites.uclouvain.be\/soumillionlab\/wp-content\/uploads\/2018\/10\/siclopp7-300x127.png 300w, https:\/\/sites.uclouvain.be\/soumillionlab\/wp-content\/uploads\/2018\/10\/siclopp7-768x325.png 768w\" sizes=\"auto, (max-width: 963px) 100vw, 963px\" \/><figcaption>Figure 1 : The SICLOPP technology. Inteinsnormally excise themselves from a protein sequence, leaving behind afull-length protein. By splitting the intein and swapping its domains, splicingof the intein leave behind two intein fragments and a circular protein orpeptide. <\/figcaption><\/figure>\n\n\n\n<p class=\"wp-block-paragraph\">In order to identify the peptides of interest from the libraries, we rely on a technique that we called &#8220;Selection by Lysis&#8221; (See Fig. 2). This let us leverage the power of selection approach and their very high throughput. After transformation in a strain expressing our target carbapenemase, the cells are grown and an antibiotic such as ampicillin is added.
